Development and functions of tissue resident NK cells
Cytomegalovirus (CMV) infects between 60 to 70% of adults. In immunocompetent individuals, infection often goes unnoticed and is cleared from most organs. However, CMV persists and eventually goes into a latent phase in the salivary glands. Reactivation of CMV in immunocompromised individuals and neonate infections can be life threatening. We are currently investigating why Natural Killer (NK) cells are poor immune effectors in the salivary glands, presumably leading to CMV persistence in this organ.
Role of phosphatases during NK cell and iNKT cell development
NK cell and NKT cell inhibitory receptors associate predominantly with the tyrosine phosphatase SHP-1 or SHP-2. Using mice conditionally deficient for these two phosphatases, we are currently examining their roles during NK and iNKT cell development. We are also investigating how these phosphatases regulate NK cell and iNKT cell immune responses.
Non-classical T cell response to viral infection
In the last 30 years significant progress has been made regarding the role and functions of CD4+ and CD8+conventional T cells. Much less is known about non-classical T cells, which include iNKT cells, MAIT cells as well as M3 and Qa-1 restricted T cells. We take advantage of a variety of tools including MCMV mutants and immunodeficient mice to determine the role of these understudied cells.
